To me it looks as if a thin layer of metal is peeling off the reverse of the coin

You're correct, US quarters are not plated as part of the minting process. People do all sorts of things to them once they leave the mint, including plating them.

Suggest soaking the coin in 100% acetone for a day or two. It's possible the 'layer' (dried crud?) could be fully or partially removed.

Peeling plating is pretty possible.

I think that is glue or something, what you are seeing is the original surface being protected by the glue.
