It could just be a trace metal present causing it. Found this article. Doesn't sound like there is any need for concern.

Looks like this one is in a PCGS holder. AU-55. The spot at 9 O'Clock could be a surface disturbance that was picked up in the photo. With the lighting moved to a different angle, it may not show up.
