What would you consider this to be? The annealing stage of the process?
If so would it affect the grade of the coin?

Not exactly sure, the photo isn't very clear to see the details. My first impression is a strikethrough. It looks cleaned with strange toning, which could be a hint of what it is.

What specifically are you asking about? The color of the obverse, the horizontal mark, the fingerprint (?) trace?

I would say some time of a struck through, grease or cloth possibly, but without sharper pics that's just a slightly educated guess.
