Run size is the number of notes in the print run that note is from. Total printed means all print runs in that particular series of star notes totaled 6,400,000, so it's not a particularly rare *. It's likely there were 2 print runs of 3,200,000. Check http://www.uspapermoney.info/serials/ they have details of print runs / serial ranges.
