Guys can you tell me what has happened here, I thought it was a stain that would come out after soaking in acetone, but it didn't.

Epoxy? Acetone does nothing to epoxy.

is that feature raised or incuse? I also see it by the 'E' too. I looks like a drop of some liquid was placed (or dropped) on this coin.

Take a tooth pick and rub it on the edge, you will probably feel a ledge, it is a drop of "something".
