I believe the letter followed by the number marks the position of the bill on an uncut sheet, and the ones on the right show the serial number of the printing plate (along with an FW if it was printed in Fort Worth.)

On a United States fifty dollar note?

If you are referring to the letter and number in the upper left hand corner, that is the plate position.

As far as I know, plate position has no bearing on the value.
