I can't tell if this is a legitimate ink error or a marker. I am hoping someone can tell me which one it is. Thanks.It's the green line above the eagle.

It look like marker to me. Maybe from someone who grabbed the wrong pen when checking to see if it was authentic?

Looks like part of a teller stamp.

When notes are stored in the vault, the teller puts them together in a strap of 100 notes, then stamps the strap with their stamp to identify who verified the strap. Often with large teller stamps, part of the stamp ends up on the first note in the strap.

Usually teller stamps are black, but this one appears to have been green ink.

It's a teller stamp. They stamp the band holding the notes together and part of the top bill to make it look more official or something.
