What is the difference between the 1976 S proof, 1976 S 40% B.U., and the 1976 S 40% Pr.?

What does BU and Pr means?

The difference is very easy to see in pictures...

This first coin is a 1976-S quarter. It is made of the clad composition that is used for quarters meant for circulation...
1976-S-Quarter

This next coin is a 1976-S 40% issue. It is 60% copper and 40% silver and it came in a special proof set. If you look at the coins devices (Washington's portrait, the date, etc.) you will see that they look "whiter" than the clad proof shown above.

(this is what the proof set looks like...)
1976-S-Quarter

The 1976-S 40% BU is also made of 40% silver and 60% copper. It is struck as a uncirculated coin and put in a special 3 coin silver mint set. This coin looks like a regular circulation issue coin...

Hope this clears things up!
