I just purchased a roll of uncirculated coins in a sealed tube. Should I leave them in there when it arrives? Or over time, will they start rubbing one another even with only occasional movement?

I actually have no experience with uncirculated coins being in tubes so another question I have is, assuming they did a perfect job placing them into the tubes and the tube is a snug fit, will just having it being sent to me through the mail already affect them?!

They are fine in the tubes and will be fine for decades.....but they are yours and if you would be more comfortable with them in 2x2's....go for it.

Actually the "tubes" would be more "airtight" for these uncirculated coins and 2x2's obviously aren't. I use both myself though. If you will have them around some humidity or heat/cold temperature changes etc. etc...then I'd just leave em' in the tubes.....assuming they have a fairly snug fit in there as you remarked.....just something to think about. Also, the more you "handle them" the more chance for them to become "less than uncirculated" if ya know what I mean ! Ha Ha.......

Leave them in the tubes. Coins stack well; only the rims should be making contact, so no rubbing or damage to the devices. I have set aside coins from circulation each year and keep them in tubes. They still look as good as the day I put them in there, going back about twenty-five years.

Ahh the rims! Okay, I understand now how leaving coins in tubes works. Especially if I probably store it horizontally, eh? Thanks for the advice all.

Quick follow up question: Are those generic two piece hard plastic circular coin holders safe to use in the long run? Not those Air-Tites with the foam, just the basic ones without foam. I've seen coins in these capsules with toning on the edges or the rims. Is it worth switching to Air-Tites for silver coins?

I say yes. I have a couple of "year sets" that I made several years ago using these kinds of holders.

I don't fully trust tubes, I put mine into zip lock bags with a dessicant pack and seal it. I've actually seen some tubes of Lincolns where some moisture got in and turned them all green.
