I'm like 99% sure this is marker.

The green looks the same as what is printed on the bill so it is possible the marks are ink splatters.
Hard to tell without the bill in hand.

This was my reply when I saw it on the CU forum.

Hard to tell. It appears to be same color as the seal and serial numbers, and the over inking of a portion of the serial number above it suggests there might have been some spatter involved. Cool find.
