That D wasn't on there when it left the mint.
Has been added after, probably by someone with a punch set

The 1950-D Nickels were lower in number, so someone was trying to turn a Philly one into a 1950-D Nickel. It was was announced that the nickels would be lesser in number that year from Denver,
Philly: 9,796,000
San Francisco: 0
Denver: 2,630,030

So a lot of rolls got saved that year. Thus the prices in BU are just a bit higher than the circulated examples.
So there were no 'S' mints that year? (Another point I had noticed until I looked it up)

An "Alchemy" Jefferson! Does a fake mint mark over the Monticello mean the coin has been turned into a 35% silver "War" alloy nickel? Nicolás Flamel would be all over this one!
ANA ID: 3203813 - CONECA ID: N-5637 Clean a coin that may be worth collecting? Please DON'T! When in doubt, leave it dirty!!

I agree its just a metal stamp into the coin and not of any value. There actually were some Jefferson nickels with the mint mark in that area. These are the 1942-1945 w/large P or S mint mark above monticello which are 35% silver. Some 1942 (denver) and no mint mark philadelphia are not silver but Denver are silver in 43,44,45. All 1943-45 had silver. https://www.coinstudy.com/1942-nickel-value.html You can see its raised unlike the one you presented.
