It looks like solvent or acid was dripped on the penny but it is very odd. Any insight is appreciated. Thanks

Definitely PMD. The glue or epoxy protected that area from toning.

For fun, try giving the coin a looooong soak in 100% acetone--might remove or reduce the feature in question. Again, only for fun.

yep, a glue of some sort that has bee non the coin for a long time - While the unprotected area toned brown, the spot under the stuff stayed relatively clean.
