Looks like carbon spotting ... localized toning on and surrounding surface contaminants. Look at the spots via 10x from a 45 degree angle. Is the surface raised at the spotting? If it is and the coin is not in a reputable slab, I'd try the acetone treatment followed by a thorough rinse.

The slab is not airtight; it won't interfere either way with the process. However, it's unlikely that the spotting will get any worse, except possibly on a *really* long timescale, measured in decades.
Do you think using something like a intercept shield slab box would help at all in keeping the spotting from spreading? or would it be just the same as putting it in a regular PCGs/NGC slab box?

They aren't airtight but they do greatly slow down the changing of the air, so while they will not stop the process of toning they will interfere by slowing it down a great deal.
