Recently pulled this from a strap of $2 notes. What's the deal with stamp that was on note and why ink stamped? Save or spend?

It is a postmark. You can see the white square where the stamp was placed.
The bill is dammaged.

Is there any value over face if bill was in better condition with stamp still on?

They weren't mailed, just stamped and cancelled. I've got one with a Michigan stamp, cancelled April 13, 1976 that my dad got for me. I'm not sure the purpose of the stamp and cancellation, though.

The first day of issue for the 1976 Series $2 was April 13, 1976. Many people had them postmarked to commemorate the occasion in a similar manner to the recent Lincoln Cent rolls and they can sell for up to $10 when CU.

Notice the postmark on this one- Jefferson Memorial Station. The USPS set up a postal station at the Arch, which is known as the Jefferson National Expansion Memorial.

unfortunately you are missing the postage stamp along with most of the postmark. The bill is worth $2.00
