i have acquired an oversized Buffalo nickel copy in an air tight container, that appears to be silver. It is dated 2001, under the buffalo and on the front under the indian head is copy # CT 1543.

Does anyone know what this is and what it may be worth?

It sounds like a privately minted bullion coin but if there is no mention of silver or ".999" it's more then likely not

Please for the future refrain from using ALL CAPS

I was thinking the same thing, or it could possibly be a copy of a 2001 Buffalo commemorative dollar.

If you have the ability to post a clear picture of both sides of it, it will help us in giving you a more definitive answer.
