I know this is probably a guess for anyone, but I'm curious if there is any significance to these 4 quarters I got as change (all at once) today with the letter C marked on them with a sharpie. I'm guessing a roll searcher did this to mark coins they've already searched, but if that's the case, why take that time and not just put some mark on the roll itself.

I have never seen anybody do that for any reason. My first guess was that they were in rolls at some place of business and somebody marked the tops of the rolls, much like banks stamp bills.
But that doesn't fly since you got them all at once in your change. All the quarters you got probably came from the same roll.
Somebody must have just been playing with their marker for no discernible reason.

I found some Halves like that the other day but there wasnt a letter on them just a red line across it. Never could figure out why someone would do that but John1's guess is a good bet.

Its a fair bet it means checked that why when the roll hunter gets them back in a later roll he will not waste time looking at them again. I mark half dollars I have checked for variety and errors. The time involved to check each coin properly is something you don't realy want to duplicate on the same coin over and over again. So I mark them with a sharpie.

Are you near San Francisco? I got some quarters marked A and B last week! Anyone see any coins marked with D?  The rolls were also marked something like K-M.

I think it could be a initial of someone that coin roll hunts myself, They mark the coins to see if they are getting the same ones back. There really no way to tell without asking the person who actually put it there so everything is just a guess but that is what I think may have been what this was

If the person has searched through, say, a thousand quarters to me it would be a colossal waste of time to write a "C" on each coin.

zhunzi, Your theory is probably correct. You have to remember that how we deposit coin is dependent on the region and your bank. Around my home turf, if you brought in rolled change, they would ask you to unroll it or charge you to unroll it because they have to recount and re-roll all change. So we turn ours in lose in boxes...no re-rolling for me :)
