I have an ACE 250 Garrett detector. Unfortunately I don't have the instructions manual. Have found a few items but am having much trouble understanding the discrimination settings. Can anyone help please?

Discrimination settings allow you to exclude some kind of metal. Just near to the coil some object that is made from a metal you want to exclude, and press DISCRIM Buttom.

If you're trying to seek for coins, ACE250 is very precise in my opinion for USA coins. The same I can say for brazilian coins (my case). ACE250 doesn´t help me because most of coins are bimetalic or are made for a mix of metals.

For the Ace 250 you can discriminate through the preset mods on the left side of the screen, or you can manually discriminate metals in each mode by hitting the discriminate button and then the eliminate button once you have the black box over the metal you wish to disc out. The Ace 250 will also save your discrimination info.
