Those look like grease/oil/water spots on the die. Not a real big thing in the collectable world. When they get to take up a greater area of the coin, they become interesting.
On the second image: The area above the "D" around the dots near the rim, that wavy area is probably where the plating has worn off the die and was starting to peel off in that area. The Canadian mint plated some of their dies to get more use out of them. I don't know how good that was. But it left many coin affected when the dies plating began to peel.
this 2008 nickel has no plating and a hole in queens head same as the one by letter e,,my local coin dealer guy told me to hold on to it to get more info,,
