Picked this coin up in a lot I bought 30 years ago back in my college days and I haven't touched it since.

I gave it an acetone bath for 12 hours and nothing changed. I have no idea if the grey streak is paint, nail polish, or some other substance.

Any suggestions appreciated. Thanks in advance

If acetone won't lick it, the only appropriate suggestion is "leave it alone." Acetone works effectively on paint and nail polish; it has to be something far stronger than that. Anything which will remove it will almost certainly do the coin more harm than good.

Soaking in Acetone for extended periods of time is useless. I suggest you try testing your Acetone. Pour some on a clean dish and allow to evaporate. If it does and no residue, should be OK. I'd try one more time with a new batch of Acetone but for only a few minutes. IF still nothing, best leave alone. May be some type of glue that at one time might have been glued into place somewhere.

Follow the polarity ladder:

1) distilled or deionized water
2) acetone (do not use fingernail polish remover)
3) xylene

That's not a bad idea. I've removed tape from many coins and the coin is usually red under it. What's interesting is that certain tapes actually work well at preserving the surface! I've been amazed a few times.
