Does the dot below ear signify a die variety?

Your coin is the JR-1 variety. The easiest way to tell is to look at the location of the tip of the 'C' in 'Ten Cents' in proximity to the talons. These photos are from PCGS, and as you can see after I cropped them, the right edge of the 'C' lines up right over the left edge of talon 2 in the scarce JR-2 variety, whereas in the JR-1 variety, the 'C' lines up with the middle of talon 2.

I don't think the dot means anything; a lot of these large diameter Capped Bust Dimes (1809-1828) have a dot between the ear and hair curl of the bust. You'd have to ask someone more knowledgeable than myself about why these dots are present, but the presence of the dot doesn't increase the value.

those dots are called centering dots. they are a reference point for laying out and engraving the die. you can find them on a lot of early american coins.

i agree with the jr-1 attribution.
