I think this could be smeared ink but it could also be pen marking. It's in a weird place and seems it could be an error. I wanted to get opinions on this I got the highest quality photo of it I could to show here. If you look at the "I" in the word FIVE there is a black mark that continues. What do you think?

Some cashiers have sharpie type special pens that check the bill to see if its real... It might be that.

Almost certainly a post-print mark of some kind.

Quite possibly a portion of a bank stamp that slightly overlapped the paper strap. It's common around these parts and the most popular color, black.
