Found in a bank roll. Rough shape, but this appears to be a struck in metal fragment. What do you think? Weighs 5.050g

Looks more like crud or some kind of Post Mint Damage ( PMD). I don't think it's an error.

interesting looking - it had the came color of the rest of the nickel. could it be clear glue or epoxy?

I agree with trying an acetone soak. Acetone is safe b/c it cannot chemically or physically alter the coins metal. Therefore using it is safe.

Is the purported fragment attracted to a magnet? Has it increased the coin's weight beyond normal parameters?

I find it a little unlikely that all the staining on Jefferson's head is contained within the anomaly.

Update: photos here are taken after soaking in acetone overnight. It looks unchanged. No part of the coin is attracted to a magnet and it weighs 5.050 grams.

Unusual for sure. It just does not look like an error.

Is the line incuse? Perhaps someone started carving on it? A Hobo nickel that went wrong?

I can't think of any possible way that could occur when the coin was struck so I am
saying it is PMD. Doesn't really matter how it happened, damage is damage.
