I received my first America the Beautiful Quarters and wonder if opening the boxes to look at the quarter rolls will diminish the value?
Thank you

Only in the fact that they would be called "unsearched" rolls. Which do bring a higher premium. There could be a major mint error in one of those rolls OR perhaps an MS70.
Other then that, there is not much difference. I allways purchase a few mint boxes. One I will keep un-opened and the others I will search for high graders or errors. The rest are stored in a pvc free plastic tube and held as BU to complete sets at the end of the run.

How does one go about getting coin rolls like this? Do you have to pay a mark up price? Or can you get stuff like this from the bank?

I would not open the boxes if you want to sell in the future. Unopened Mint boxes do have a premium over regular mint wrapped rolls. They also have a time stamp on the side of the box which may provide information to variety searchers that track time frames for possible error finds.

Lukky, you may be able to find uncirculated mint state rolls from your bank at face value but we are talking about the US Mint's (website) Products that have special wrappers and are 100% guaranteed to house uncirculated coins.
