The mark is on the outside. I cropped it out, but there are markings on the cardboard too; it looks like the entire thing was kept in some sort of money clip.
I tried to push the coin to side a bit by pushing on the plastic next to the coin, and I fear that it might of left a mark on the coin as well but I couldn't get a clear enough view to know for sure.

What are those white stripes on the eagle ? It looks like there was some kind of impact and pieces of metal fell off.

Ouch, if the black stripes are on the coin, that is a real big distraction. The OP says it is on the holder, so I will take their word for it. It does appear to continue past the coin on the plastic. This high grade 1829 Capped Bust half dollar is the O-115 variety that has a rarity of R-1, 1000+ known examples and commonly found. The stars, hairline, wings, arrowheads, and talons are all strongly struck, making this variety a nice type coin for the series. This coin has nice rims, and a very original toning pattern. This one is AU-58, because of the slight darkness on the high points of Miss Liberty. That is minor rub, keeping this coin from being uncirculated. I put a value of $600 on this coin.

AU58, but a MS grade would not be a surprise.

First, I agree with the AU-58 grade and upthread comments. I wonder if the black mark could be the residue from an old rubber band?

AU58, would not be surprised at a MS62/63, hard to decipher wear vs slightly weak strike sometimes.

Are you deliberately ignoring this?

I'm going to be the oddball here and say MS-63. Needle sharp. Lots of luster. Great eye appeal. Zero distractions. I'm not sure where you all are seeing the rub. This coin never saw circulation.
Slirk, those lighter areas are the high points on the reverse and the tarnish is lighter in color there. This is not a problem. It is not damage and not distracting in my opinion. I've collected bust halves for 30 years and these areas often show flatness due to the metal not flowing up to the highest points of relief and/or making contact with the surfaces it has been laid on.

Thanks for the explanation. It was important to me.

looking at this coin again I can say it could be MS also. my only concern is that we cant see the full cheek due to the marker. toning makes it a little more challenge to see minor circulation wear. if it makes the MS grade i'd put it at MS62. if it comes back as a 62 then that more than doubles its value from a 58 at about $1600
