In general you won't find large dollars at the bank unless someone has just turned some in. If you ask you might get lucky, but they won't be mixed in with a dollar box. I have heard that banks can order large dollars from the federal reserve, but I haven't heard of a bank actually doing that. I imagine there must be a lot sitting in a vault somewhere.
