I believe the upper right number is the note's Treasury serial number while the lower left number is the individual bank serial number. The BEP eventually did away with this numbering system later on towards the end of the large size National bank note issuance.
By the way, the 4 digit brown number (below the Treasury serial number) is the bank's charter number.
