I'm looking to make a large profit from selling coins. My main areas are War Nickels, IHCs, V nickels and Buffalos.

T-BOP, be nice.
If you can get those coins for face value and sell them for 50¢ a piece, that would be a 1000% profit, which would indeed be large.
But to answer your question more seriously, there is no particular market where you can just walk in (or use online) that will guarantee you will get a "retail" price.
Also, there is no particular market that will guarantee you a large profit. Profit is dictated by sales price and cost.

The best way to get retail is to sell direct to the collector,the bay-Craigs list-coin club John1

Maximum visibility gets the best prices, and there is no larger coin market than ebay. You have to pay fees and accoint for shipping (minimum is $2.66, but you can fit a LOT of coins in a flat rate box!) But as long as individual coins are nice and/or grouped in big enough lots to be enticing, you will make more on ebay than at a LCS and won't have to waste an entire day at a flea market table.

Only Littleton and similar firms can manage that feat. The rest must use ebay or the likes to get the "real" market price.

 with john1. You would have to sell to a collector to get the highest prices for your coins and e-bay is your best bet .

ebay minus fees and expenses is your best bet. If you really had a lot to sell, you could try renting a table at a local coin show.
