Will the missing R in LIBETY increase my circulated 1916 Wheat cent value?

Would have to see pictures. The R could be missing for any reason from post mint damage, to a Struck Through Grease, to a more major strike through, a lamination, or possibly something else entirely. I would say most likely no increase in value unless it's a major strike through, lamination, or something else fairly significant.

@nj, first welcome to CCF. Second, while I agree with everything that @sc said, I note that even if the missing letter is from grease, that isn't enough of the design missing to really affect the value. Usually, more of the design elements (think whole words) need to be missing before you get much of a bump in price due to a grease-filled die.

Didn't mean to suggest a Struck Through Grease would add value, just if it were struck through something more interesting.

Good point @SC. Now that I re-read your original post, I actually agree with everything you said.

Just the over 80%+ grease filled examples might have a premium. It all depends on how bad someone wants them. Single devices is not a big deal:

Usually a missing letter or number are not actually missing. most times you can actually see the missing letter or number using a scope or magnifying glass or even turning the coin a certain way. This is usually due to the reasons listed above or highly polished die. Depending on what the cause of the missing letter or number will determine the value. If you post a picture we might be able to at least point you in the right direction if not give you a definitive answer.

If the missing letters are from damage, it is just a face value coin. Damage is damage. Just look at my car. A wrinkled fender, does not increase value.

Lol. Wrinkled fender. I think that's (one) of the reasons my wife wants to turn me in for a new model.
