What causes this on a quarter anyone know?

If found in circulation, something was added to the coin. If from an original role, something affected the stock material. Neither are a plus for the coin. Just an imperfection of the metal or some event in circulation bonded to that area. Again, not a plus the the coin.

Who knows what it is, some kind of stain. Didn't happen at the mint.

Looks to me like a patch of crud is adhering to the coin's surface. Is the feature raised above the surface, or perfectly flush?

Hard to say unless you were there at the time. It could be anything that
Would cause a stain. Whatever it is, it is PMD

It is not flush it looks like a layer is gone...I thought it was interesting positioning better that the poopy horse on Wyoming horses!
