I'm in with staining. Might try giving the coin a long soak in 100% acetone--might reduce or eliminate the stain.

Looks like a stain across the area you are looking at. If the field is level then it is a stain.

If it is cladding issue, there should be more red color present. It may have been a struck through a piece of cladding debris on that area and has fallen off if the color is not there.
Normal weight is expected on the struck through debris issue.
Slightly under weight on the cladding loss on that area.
