The black spots are irremovable by any means without destroying the patina. Somebody has already attempted conservation on this coin, probably mineral oil (i.e. Coin Care), sorry to say it would not slab. That's the only reason I will continue....
That said....I do see some remaining verdigris below the portrait near the rim and it looks like a spot on the reverse between the AT in STATES. For those I would put a little on a toothpick (I prefer STIM-U-DENT plaque removers, pointer) and carefully apply small drops over the verdigris. Let it stand for about 5 minutes and gently tap and/or rub to remove the verdigris. Then dab off the excess with a dry towel and inspect. Repeat if needed, then allow the coin to dry for a minimum of 24 hours before putting in a holder.
Not sure your experience level with conservation, but you might want to practice on some junk coins before you dive in. Also, be VERY careful not to rub off the patina! I've accidentily gotten too aggressive with my rubbing and removed patina before....it's a sad moment. LOL
