I will be relocating and need to transport my coins a long distance by car(to a secret location of course). My concern is I have many tubes that are not completely full and don't want the coins to move around and damage each other. Any recommendations as to what I could put inside to keep them from rubbing (such as cotton), should I transport vertical or horizontal, or am I crazy and over-thinking this?

Vertical would be best, stuffing it with cotton, bubble-wrap or something soft that in itself won't damage the coins but will prevent them from moving around should work best. And no, you're not overthinking this.

Not a problem at all, if your uncirculated standard coins are bullion only. Follow the suggestions already mentioned.
Low value coins below EF can be transported in bulk, if that is the way you already store them.
The problem is already solved for those coins currently in albums.

Make sure that your whole collection is in plain anonymous looking corrugated boxes, so that undesirable eyes don't see the contents, during your move.

I am having my house renovated soon.
My safe, although it is bolted to the floor inside a cupboard, will nevertheless be covered by a plain looking corrugated cardboard box. The key will be off the premises, miles away.

Sel's advice is sound as is zxcccx's.

If you have to spend a night or two in a hotel, bring it ALL in to your room and don't spend much, if any time with it out of your sight. It may be a pain to drag it all in and then back out the next day but it's worth it.

Cotton should be fine. I was also thinking facial or toilet tissue, most people have that in their house already.

If you have styrofoam packing peanuts lying around, those work well in stuffing partly filled rolls as well as the other kinds of packing materials mentioned above. Another note is not to put all of your coins in one box, it gets very heavy quickly and if people see you struggling with the box, they will think you have gold bars inside. A small cardboard box for your most prized and/or expensive coins that can be hidden inside luggage or a large duffel bag is another strategy. Good luck on your move.

I would use small zip-lick baggies with enough air to take up the slack in the tubes.
