Don't clean it!... To late, you already did it. Don't clean coins.
That spot could be anything, environmental damage, (post mint damage) a chemical of some type. I wouldn't necessarily say intentionally marked. That spot is most likely baked into the metal/coin and cannot be removed. Put it in a 2x2 flip and just leave it alone. It's still a nice "silver" coin.

Just this once I used a mild soap and just my fingertip. You're so right about not cleaning.

@jim0815, yes pretty hard to say what it is. Before you put it into a 2x2, consider giving it a dip in acetone. If that mark is in fact ink, that should remove it.

@Spence, Thank you for the advice. I know to use pure acetone and not nail polish remover. You guys on this site are great and I thank you.

Well, the pure acetone did absolutely nothing to the mark. Still there.
