I think she is just showing off her perfumed glove. (I can find paintings of her on the internet where she wears no gloves, gloves on both hands, glove on right hand, or glove on left hand, so nothing wrong with her hands.)

Perfumed gloves were very fashionable among royalty and upper aristocracy in the 1500s and there are many paintings of royal persons with gloves, often just holding one or a pair in one hand. For instance, Elizabeth I of England is often portrayed holding gloves in one hand. Or just look at the painting of Sigismund II in the post by tdziemia above...

Making leather gloves in the 16th century involved treating them with various obnoxious organic substances (I will not be more specific, use your worst imagination... ), leaving them with a rather rancid odor. They had to undergo a long and complicated and costly treatment with herbs, flowers, and other fragrances before being presentable in nobler surroundings.

Today I have the same coin as @Petrus.
There was quite a bit of innovation in denominations during this Sigismund's reign. The 2 denar, and 4 groschen coins we have seen here the last three days were minted for the first time under Sigismund II (also the 2 groschen).

a Bourgondische Kruisdaalder (Burgundy taler) minted in Antwerp in 1568, which the familiar "klop" of the rebels in 1572
