Looks to me like a small stike thru of a piece of something that has since fallen out.

Try a toothpick on this. It is usually clear/white grease. That has been my experience. sometimes it is elmers glue. But most of the time a toothpick removes it.

I looks like the area was indented into the coin. Is it indented or is it sticking up?

It appears to be clear glue and will come off with acetone.

I feel it is the effect of having been struck through something. As mentioned above, it could have been a fragment of metal that has since fallen away.

Probably struck through a fragment of metal, it is always nicer when it crosses the devices rather than just in a field.

It's not glue, don't pick at it with a toothpick, that'll just scratch the coin. Like Bill et al have said, it's a struck through.
