Looks like glue as it is clear. If it were hot metal, we would not be able to see through it.

Looks like some type of clear glue. Acetone for 24 Hours should help or tooth pick your choice.

This looks like a Grease Filled Die strike to me. It is possible to have a very shallow lamination on the clad layer of a coin, but it looks a lot different. The depth of the area in question is deeper than the clad layer is thick. I also see what looks like damage on the coin.

Do you know the weight of the coin?

Edit: Is the area of the anomaly raised or incuse. I thought it was incuse from the images, but the other member's answers make me think otherwise.
