Looks like a bit of dried adhesive that has now darkened. You could try to soften it up in an acetone bath or just spend it.

I actually think it's normal wear. I think these nickels are 2/3 copper. I think it's the copper coming thru.

Alright thanks fellas just didn't think it was the copper coming through since I have searched so many nickels and never seen it before on much more worn coins but that is why I ask the experts because I am far from that. Thanks again.

The test would be to see if the coins weight is heavy. That would probably be detected. Something added to the coin, would add weight.
5g is the correct weight.

I'd say adhesive has stained the surface. Nickels have never been made as clad or plated coins.

Yeah must be some adhesive because the coin weighs 4.95 grams. Well gave me something to post. At least the box of nickels in came in was one of the best I have opened. Thanks everyone for the help!

The metals are a mixed alloy/ The coins are not copper coated with nickel. So wear cannot reveal a copper layer/core underneath.

Look what I just found. Looks very similar. I don't see any adhesive or stain. It may have been affected by some type of chemical or acid. It still looks like normal wear to me. It's the high point on the coin. The most likely spot.
